Effect of iron on the anticancer activity of in vitro photodynamic therapy using Photolonᵀᴹ

Photodynamic therapy (PDT) is a minimally invasive cancer treatment applicable across various cancer types and stages. This study aimed to enhance PDT efficacy by evaluating the effect of iron on PhotolonTM-based PDT in vitro. CT26 and HeLa cell lines were treated with PhotolonTM (0-12 μg/mL) as the photosensitizer using irradiating laser (660 nm, 3 J/cm2) and VenoferrumTM (0-12 μg/mL) as the iron source. Cell viability was measured using the MTT assay after 24 h, while intracellular reactive oxygen species (ROS), lipid peroxides, and 8-OHdG contents, which are indicators of oxidative damage, were measured after 6 h of laser irradiation. After 24 hours of in vitro PDT, photocytotoxicity in both cell lines increased in a concentration-dependent manner at PhotolonTM concentrations 4-10 μg/mL. Co-treatment with iron further enhanced PDT-induced cytotoxicity compared to PhotolonTM alone. Similarly, ROS, lipid peroxides, and 8-OHdG levels were elevated in the PhotolonTM plus iron group relative to PhotolonTM alone. These findings suggest that iron increased photocytotoxicity in PDT using a PhotolonTM by increasing oxidative cell damage due to the increased production of intracellular ROS with iron.
