Human pain research is ethically barred from deliberately inducing tissue injury, making carefully controlled animal models indispensable for mechanistic discovery and early-stage drug screening. These preparations allow strict control of genetics and environment, invasive mapping of neural and molecular pathways, and pre-clinical testing of safety and efficacy. This review concisely profiles the main animal paradigms that mimic inflammatory, neuropathic, visceral, muscle, post-operative, cancer-related and ischemic pain. For each, we outline standard induction methods, key pathophysiological drivers and the time-course of hypersensitivity. Reflexive assays (e.g., von Frey, Hargreaves, tail-flick) and non-reflexive measures (e.g., formalin phase II, grimace scale) are matched to the models they best interrogate. Thoughtful model-to-hypothesis matching, meticulous methodology and transparent reporting remain the keys to maximizing the clinical relevance of preclinical pain research.
