국문 초록

본 연구는 레몬그라스(Cymbopogon citratus) 에탄올 추출물(ethanol extract, LGEE)과 에센셜 오일(essential oil, LGEO)의 Streptococcus mutans에 대한 항균 효과를 분석하고자 하였다. 항균 효과 분석을 위해LGEE는 125 mg/mL, 250 mg/mL, 500 mg/mL를, LGEO는 5%, 10%, 20%, 30% 농도를 사용하였다. 또한 디스크확산법(disk diffusion method)을 이용하여 clear zone의 형성 정도를 측정하였고, 최소억제농도(minimum inhibitory concentration, MIC)와 최소살균농도(minimum bactericidal concentration, MBC)를 확인하였으며, 배양시간별 성장억제 효과를 분석하여 다음과 같은 결과를 확인하였다. LGEE는 125 mg/mL에서 14.89 mm의 clear zone을 형성하였고 LGEO는 5%에서 18.56 mm를 보여 LGEO의 항균 효과가 더 강했고, 두 시료 모두 농도 의존적인 경향을나타냈다. LGEE의 MIC는 0.8 mg/mL에서, LGEO는 0.6~1.2%에서 나타났고, LGEE는 1.6 mg/mL에서, LGEO는1.2%에서 각각 집락이 형성되었다. 성장억제 효과는 대조군 0.603 대비 LGEE가 125 mg/mL에서 0.144, 250 mg/mL에서 0.132, 500 mg/mL에서 0.123인 것에 반해 LGEO는 대조군 0.628 대비 5%에서 0.128, 10%에서 0.117, 20%에서 0.108, 30%에서 0.105를 보였다. 결론적으로 LGEE와 LGEO 모두 S. mutans에 대한 유의한 항균력을 보였고, 두 시료의 활성 비교에서는 LGEE보다 LGEO가 더 강한 항균 활성을 나타냈다. 추후 연구에서는 어린이 음료의 첨가에 용이한 레몬향의 소재인 레몬그라스 열수 추출물의 항균 효과를 검증하고 치아우식증 예방을 위한잠재적인 기능성 소재로서의 다양한 접근을 위한 시도가 필요할 것으로 생각한다.

영문 초록

This study examined the antibacterial effects of a lemongrass (Cymbopogon citratus) ethanol extract (LGEE) and essential oil (LGEO) against Streptococcus mutans. The antibacterial effects of LGEE were tested at concentrations of 125 mg/mL, 250 mg/mL, and 500 mg/mL. LGEO was analyzed at doses of 5%, 10%, 20%, and 30%. The results were obtained using the disc diffusion method, minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC). The growth inhibition effect was measured 3, 6, 12, 24, and 48 hr after treatment. The growth inhibition effect of LGEE and LGEO was 14.89 mm at 125 mg/mL and 18.56 mm at 5%, respectively. Hence, the antibacterial effect of LGEO was stronger than that of LGEE. The MIC values of LGEE and LGEO were 0.8 mg/mL and 0.6-1.2% (v/v), respectively. The MBCs in LGEE and LGEO were 1.6 mg/mL and 1.2% (v/v), respectively. The growth inhibitory effects after 48 hr of incubation were 0.144 at 125 mg/mL, 0.132 at 250 mg/mL, and 0.123 at 500 mg/mL for LGEE compared to 0.603 for the positive control. In contrast, LGEO was 0.128 at 5%, 0.117 at 10%, 0.108 at 20%, and 0.105 at 30% compared to 0.628 for the positive control. As a result, the LGEE and LGEO exhibited significant antibacterial activity against S. mutans. A comparison of both agents revealed LGEO to have more potent antibacterial activity than LGEO. Lemongrass has been used as a tea and a spice in food, especially lemongrass oil, which has diverse uses because of its versatility. Future studies should examine the antibacterial effects of a lemongrass water extract, a lemon-like flavoring ingredient that can be easily added to children’s beverages, and different approaches to prevent dental caries.
