Hesperidin과 quercetin의 복합처리가 비알코올성 지방간에 미치는 영향

국문 초록

헤스페리딘과 퀘르세틴은 항산화와 항염증 작용이 있는 것으로 널리 알려져 있다. 본 연구는고지방식이 섭취로 비만이 유도되는 C57BL/6J mice에서 헤스페리딘과 퀘르세틴의 단독처리 및 복합처리가 비알코올성 지방간을 개선하고 간에서의 염증 억제 효과가 있는지를 조사하였다. 마우스는 무작위로 고지방식이 섭취 대조군(Con), 헤스페리딘을 처리한 고지방식이 섭취군(Hes), 퀘르세틴을 처리한 고지방식이섭취군(Que) 및 헤스페리딘과 퀘르세틴을 복합처리한 고지방식이 섭취군(Hes+Que)으로 분류하였다. Que 과 Hes+Que의 간 무게는 통계적으로 유의하지 않지만 Con에 비해 감소하는 경향이 관찰되었다. 그리고Hes, Que 및 Hes+Que는 Con에 비해 몸무게와 간세포의 지질축적이 현저히 감소하였다. 간세포에서 염증 표지 유전자의 발현을 분석한 결과, Con에 비해 Hes, Que 및 Hes+Que는 염증 인자가 감소하였고 항염증 인자는 증가하였다. 이러한 연구결과들은 헤스페리딘 및 퀘르세틴의 단독처리보다는 복합처리가 더효과적이었다. 따라서 본 연구는 헤스페리딘과 퀘르세틴이 비만으로 유도된 비알코올성 지방간과 간세포의염증에 대해 억제효과가 있으며, 특히 헤스페리딘과 퀘르세틴의 복합처리가 각각의 단독처리에 비해 간 기능을 더 효과적으로 개선함을 시사한다.

영문 초록

Hesperidin and quercetin are widely known to have antioxidant and anti-inflammatory effects. It was investigated whether single or combined treatment of hesperidin and quercetin improve non-alcoholic fatty liver and suppresses hepatic inflammation in high fat diet-induced obese C57BL/6J mice. Mice were randomly divided into High fat diet-fed mice group (Con), hesperidin-treated high fat diet-fed mice group (Hes), quercetin-treated high fat diet-fed mice group (Que), and hesperidin and quercetin-treated high fat diet-fed mice group (Hes+Que). The liver weights of Que and Hes+Que decreased, although not statistically significant. In addition, Hes, Que and Hes+Que significantly decreased body weight and hepatic lipid accumulation compared to Con. As a result of analyzing the expression of inflammatory marker genes in hepatocytes, Hes, Que, and Hes+Que decreased inflammatory factors and increased anti-inflammatory factors compared to Con. These research results showed that combined treatment with hesperidin and quercetin was more effective than single treatment. Therefore, this study suggests that hesperidin and quercetin have an inhibitory effect on obesity-induced non-alcoholic fatty liver and hepatocyte inflammation, and in particular, the combined treatment with hesperidin and quercetin improves liver function much more effectively than treatment with either alone.
