Background/Objectives: Recent industrialization has led to a dietary shifts and increased environmental pollution, both of which place significant stress on the human body and contribute to the onset of chronic diseases. Consequently, there is growing interest in safe natural materials containing a variety of bioactive compound that may help in the prevention and treatment of chronic diseases. We aimed to evaluate the antioxidant activity and anti-inflammatory effects of Lycium chinense Miller leaf ethanol extract and partially chlorophyll-removed ethanol extract(LCML and LCML with CR).
Methods: The antioxidant activities of Lycium chinense Miller leaf extracts were assessed by measuring total polyphenol and flavonoid contents and radical scavenging activities. We also examined the anti-inflammatory effects of Lycium chinense Miller leaf extracts on lipopolysaccharide (LPS)-induced inflammation in RAW264.7 cells. The effects of these extracts on nitric oxide (NO), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) releases were measured in LPS-stimulated RAW264.7 cells.
Results: Total polyphenol content, 2,2’-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity and ferric iron reducing antioxidant power (FRAP) values were significantly higher in LCML with CR than in LCML. The cytotoxicity assay using RAW264.7 cells showed no toxicity at all concentrations(31.5~1000 ㎍/mL). NO, TNF-α and IL-6 release from LPS-stimulated RAW264.7 cells treated with these extracts decreased with increasing extract concentration. TNF-a and IL-6 release tended to be higher in cells treated with LCML with CR than those treated with LCML.
Conclusion/Implications: The ethanol extracts of Lycium chinense Miller leaf and LCML with partially removed chlorophyll may serve as functional materials with antioxidant and anti-inflammatory activities.
