Yoensan Ogae as Korean black body fowl that has been designated as a natural monument (registration number 265) is genetically distinguished from other species. The peptides such as Ala-Pro-Ala-Pro-Ala-Pro-Ala-Pro expressing the antioxidant activity produced from the meats. The bioactive peptides must retain active during digestion and absorption before entering the blood stream. Therefore, this study was to evaluate the degree of release of peptides during passing the gastrointestinal environment. To manufacture the liposoms, soy lecithin, and peptides were hydrated 10 ml distilled water containing peptides (3%, w/v). Hydration was performed for 1 h at room temperature under a nitrogen atmosphere. Then, the mixture volume was made up to 100 ml with 3% (v/v) glycerol in phosphate-buffered saline. The pH of the mixture was adjusted to 7.4 and heated to 65°C with continuous shaking for 60 min under a nitrogen atmosphere. The crude liposomes were homogenized using a high pressure homogenizer, microfluidizer, at a pressure of 17,000 psi (117.2 MPa) for 5 cycles. Chitosan-coated (CH-coated) liposomes were prepared by mixing the liposome suspensions with an equal volume of chitosan solution. In order to assess the stability and protective effects imparted by the CH coating layer, coated and uncoated liposomes were incubated in simulated-gastric fluid and simulated intestinal fluids. The peptides were analyzed by Bicinchoninic Acid (BCA) Protein Assay Kit. The Characterization of chitosan coated liposomes containing peptides were analyzed by a SEM and Zeta potential measurements. The loss of peptides were found from the both cases. CH-uncoated liposomes released peptides about 40%, but CH-coated liposomes about 20% in simulated-gastric fluid. CH-uncoated liposomes released peptides about 80%, but CH-coated liposomes about 40% in simulated-intestinal fluid.
