국내 사육 해달로부터 분리한 plesiomonas shigelloides의 생물학적 특성 및 병원성에 관한 연구

국문 초록

수족관에서 사육 중 폐사한 해달 1두와 설사증을 보인 해달 3두로 부터 원인체를 분리 동정하고, 분리균주의 생화학적 특성과 병원성을 규명하였던 바 다음과 같은 결론을 얻었다. 심한 설사, 탈수, 식욕부진 등의 증상을 보이며 폐사한 해달 1두는 실질장기에서 육안적으로 심한 출혈소견이 관찰되었으며, 폐사한 해달의 각종 장기와 심한 설사증을 유발한 해달 3두의 분변 및 수족관의 물로부터 원인체를 분리하여 생화학적 성상 검사를 실시하였던 결과 분리주 18주 모두는 Plesiomonas shigelloides로 동정되었다. 분리주 18주에 대한 약제 감수성 시험을 실시한 결과 cephalothin, nalidixic acid, doxycycline, erythromycin 및 tetracycline에는 감수성이었으며, olendomycin, clindamycin, trimethoprim, sulfamethoxazole, lincomycin, streptomycin, triplesulfa, novobiocin의 약제에는 저항성 이었다. 분리한 P. shigelloides는 β-hemolysis를 산생하였고, Congo red 결합능이 있으며, Vero세포에는 세포독성을 나타내지 않았으나 Chinease hamster ovarian cell에서는 세포독성이 있었다. 또한 마우스에 대한 LD₅₀는 5×10⁷ CFU이었고, plasmid분석에서 large plasmid를 보유하고 있음이 확인되어 분리주는 병원성이 있는 것으로 생각되었다.

영문 초록

This study was carried out the isolation and identification of microorganism from tissues and diarrheal feces to investigate a cause of dead sea otter in Korea. We tested the antimicrobial susceptibility by disc diffusion method, and also examined the production of hemolysin plasmid analysis, Congo-red binding, 50% lethal doses to mouse, cellcytotoxic assay to determine the pathogenicity of the isolates. Four cases of Plesiomonas shigelloides infection were defined by isolation of the microorganism from a dead sea otter with diarrhea and bacteremia, and 3 sea otter with diarrhea. The eighteen isolates from sea otter and aqarium water were identified as P. shigelloides by biochemical tests. All 18 isolates of the organism were suseptible to cephalothin, nalidixic acid, doxycycline, erythromycin and tetracycline, but resistant to olendamycin, clindamycin, trimethoprim/sulfamethoxazole, lincomycin, streptomycin, triplesulfa and novobiocin. All isolates of the P. shigelloides produced a hemolysin on sheep blood agar, bound Congo red, and produced elongation of Chinease hamster ovary cells, but did not produce a cytotoxin on Vero cells. The P. shigelloides isolates were possed large plasmid, and the median 50% lethal dose in mouse of the strains was 5×10⁷ CFU. These results suggest that the isolates might be condidered as enteropathogens.
