꾸지뽕나무 뿌리 추출물이 C2C12 세포의 mTOR 매개 근육 분화에 미치는 영향

국문 초록

골격근육은 몸의 자세를 유지하고 지탱하게 해주며 골격근육의 유지를 위해서는 지속적인 근육세포의 분화가 필요하다. 이번 연구에서는 꾸지뽕뿌리 추출물을 근아세포 C2C12 세포에 처리하여 꾸지뽕뿌리 추출물에 의한 근육세포 분화 효과와 그 신호전달 과정을 알아보고자 하였다. 꾸지뽕뿌리 추출물은 C2C12 세포에서 IGF2-ME 활성화를 증가시켰으며, 이러한 증가가 mTOR 신호전달억제자 rapamycin에 의해 억제됨을 증명함으로써 mTOR 신호전달경로에 관여함을 증명했다. 꾸지뽕뿌리 추출물은 C2C12 세포의 근관 형성, 융합, 분화 지수를 유의미하게 증가시키고, 초기분화인자 MyoD 발현을 증가시키며 PAX7 발현을 감소시켰다. 이로부터 꾸지뽕 뿌리추출물이 근육세포의 분화를 유도하고 그 분화에 mTOR 경로를 매개로 한 신호전달과정이 관여함을 알 수 있었다.

영문 초록

Skeletal muscle differentiation is essential to maintain proper posture and long-term functional stability. Understanding the mechanisms regulating myogenic differentiation can facilitate the development of effective therapeutic approaches for muscle degeneration. Therefore, this study aimed to explore the effect of the Cudrania tricuspidataroot extract (CRE) on the myogenic differentiation of C2C12 myoblasts, focusing on the mechanistic target of rapamycin kinase (mTOR) pathway. CRE enhanced the insulin-like growth factor 2-muscle-specific enhancer activity,which was suppressed by rapamycin, thereby confirming mTOR pathwayinvolvement. Additionally, CRE significantly promoted myotube formation, fusion, and differentiation and modulated the levels of key transcription factors, myogenic differentiation factor (upregulated) and paired box-7 (downregulated), indicative of early-stage myogenesis. Overall, CRE promoted mTOR-mediated differentiation of C2C12 myoblasts, acting as a novel therapeutic candidate for muscle degeneration and associated disorders.
