감마방사선 조사 후 버섯류 - 마늘 혼합 추출물이 염증 반응 및면역 회복에 미치는 영향

국문 초록

본 연구에서는 버섯 4종과 마늘에서 유효 성분을 추출하여 적절한 배합 비율로 혼합하고 이를 실험용 마우스 모델에 경구 투여한 후, 감마방사선 노출에 대한 염증 반응 및 면역 회복에 미치는 영향을 조사하였다. 감마방사선에 노출 후 시료 추출물을 투여한 실험군에서 백혈구 수와 면역글로블린G 생성량을 확인하였고, 유전자 발현 분석및 유세포 분석 등을 통하여 면역세포활성 및 세포주기 등을 조사하였다. Mixed-M/G2 추출물을 투여한 실험군에서 면역 조절에 관여하는 사이토카인인 IL-10과 IL-2의 발현은 증가하였고, 염증 반응을 촉진하는 사이토 카인인 TNF-α와 IL-6의 발현은 억제되었다. CD11b 및 CD8의 세포비율은 각각 62.99%와 19.88%로 대조군에비하여 높은 비율의 면역세포군을 나타내었고, 세포주기 분석에서 세포비율이 G0/G1 단계에서 76.59%로 증가 하고, S 단계에서 12.52%로 감소하는 결과에 의하여 정상수준으로의 세포주기회복을 확인하였다.

영문 초록

In this study, active ingredients were extracted from four types of mushrooms and garlic, mixed in an appropriate ratio, and orally administered to experimental mouse models to determine their effects on the inflammatory response and immune recovery after γ-radiation exposure. The white blood cell count and immunoglobulin G levels were determined in the extract-administered experimental group after γ-radiation exposure. Additionally, immune cell activity and cell cycle were investigated via gene expression analysis and flow cytometry, respectively. In the mixed-M/G2 extract-administered experimental group, expression levels of the cytokines involved in immune regulation, interleukin (IL)-10 and IL-2, were increased, whereas those of cytokines promoting the inflammatory response, tumor necrosis factor-α and IL-6, were decreased. The percentages of CD11b and CD8 cells were 62.99 and 19.88%, respectively, with a higher proportion of immune cells in the experimental group than in the control group. Furthermore, the percentage of cells in the G0/G1 phase increased to 76.59%, whereas that of cells in the S phase decreased to 12.52%, confirming cell cycle recovery in the extract-administered experimental group.
