The objective of this study was to develop a new approach for fabrication of 3D printed tablet containing active pharmaceutical ingredients (APIs), exploiting the hot-melt extrusion (HME) coupled with 3D printing technology.
Theophylline was used as a model drug, and hydroxypropyl cellulose (HPC), hydroxypropyl methylcellulose (HPMC) and stearic acid (SA) were used as a carrier or plasticizer for the preparation of drug-loaded filaments. Four different compositions were hot-melt extruded, and three of them available for 3D printing showed a zero-order sustained release over 9 hours. Overall, 3DP technology coupled with HME could be a novel method to produce controlled release tablets providing the advantage of simplicity and versatility than the conventional method.
