The purpose of the present research was to examine fetal resorption and the eventual termination of pregnancy resulted from the teratogenic and toxicological effects of PMSG In pregant rats when injected on days 12, 14 or 16 of pregnancy. And, in intact rats treated with PMSG on days 6, 9 or 12, this research evaluated whether additional daily injections of progesterone from 6, 9 or 12 to the maximum of days 15 prevent a fetal resorption and the eventual termination of pregnancy induced by PMSG treatment alone.
The results was obtained as follows :
1. Treatment of rats with PMSG on days 12, 14 or 16 of prgnancy resulted in the decrease of average litter size and weight, average fetal weight and crown-rump length, and maternal body weight on days 12, no effect obtained on days 14 or 16.
2. Treatment of rats with PMSG on days 12, 14 or 16 of pregnancy had no effect on the fetal body water, the average kidney and liver weight as well as percent water for these organs, and maternal liver weight and percent water.
3. Treatment of rats with PMSG on days 6, 9 or 12 of pregnancy was able to induce fetal resorption and the eventual termination of pregnancy, but it was inactive on days 14 or 16. And, treatment of rats with PMSG by administering progestrone daily did not succeed in maintaining pregnancy.
4. Treatment of rats with PMSG on days 12, 14 or 16 of pregnancy had no effect on the concentration of sodium and potassium in the conceptus and maternal plasma. And ratios of sodium to potassium of the conceptus was much lower than those of fetuses with head abnormalities induced by a number of teratogens.
5. As the treatment of rats with PMSG on days 12, 14 or 16 of pregnancy was shown to produce no apparent adverse teratogenic or toxicological effects, it is suggested that fetal resorption and eventual pregnancy-terminating effects of PMSG in pregnant rats did not result from the teratogenic and toxicological effects or the failure of progestrone.
