열소작술을 이용한 흰쥐 외안근 섬유화 모델

국문 초록

목적: 쥐에서 외안근 섬유화를 유발하는 데 있어 열소작술을 이용한 열손상의 효과를 평가하고, 이상적인 외안근 섬유화 연구 모델을 설계하고자 하였다. 대상과 방법: Sprague Dawley 흰쥐 12마리의 24안들을 무작위로 2개의 군으로 나누었다. 실험군은 상직근을 노출시키고 상직근의 부착부로부터 후방으로 2 mm 떨어진 곳에 1초간의 열손상을 2번 가하였다. 대조군은 열손상을 제외하고 동일한 방법으로 조작하였 다. 2주 경과 후, 24안 모두를 적출하였고 Hematoxylin and eosin (H&E) 및 Masson s trichrome (MT) 염색을 시행하였다. 결과: H&E 및 MT 염색을 통해 열소작술을 시행한 군에서 통계적으로 의미있게 염증반응과 섬유화가 증가된 것을 확인할 수 있었다 (각각 p=0.002, p<0.001). 결론: 조직학적 결과들을 통해서 쥐 모델에서의 열소작술이 효율적으로 외안근 섬유화를 유발할 수 있음을 확인하였다. 이 모델은 상직근 섬유화를 유발하는 데 있어 효과적이고 사시수술 후 섬유화 발생을 연구하는 데 유용할 것으로 생각된다. <대한안과학회지 2018;59(5):478-483>

영문 초록

Purpose: To evaluate the effect of cauterization on extraocular muscle (EOM) fibrosis in rats, and to develop a novel EOM fibrosis model. Methods: Twenty-four eyes of 12 Sprague Dawley rats were assigned randomly to two groups. We exposed the superior rectus muscle (SRM) and performed thermal injury 2 mm behind the insertion site of the SRM using a cautery device in the experimental group. The thermal injuries were performed twice for 1 second, for a total of 2 seconds. In the control group, the same procedures except the thermal injury were performed. Two weeks after surgery, all eyes were enucleated and stained with hematoxylin and eosin (H&E) and Masson s trichrome (MT). Results: Staining with H&E and MT showed that thermal injury significantly increased inflammation and fibrosis in the experimental group (p = 0.002, p < 0.001, respectively). Conclusions: Thermal injury using cauterization effectively induced fibrosis of EOM in the rat model. This simple model was effective in inducing fibrosis of SRM and will be useful for studying postoperative fibrosis after strabismus surgery. J Korean Ophthalmol Soc 2018;59(5):478-483
