In order to raise the efficiency of callus induction and plant regeneration, somatic tissues of tall fescue(Festuca arundinacea L.) such as leaf and root from 7 days old seedling, mature embryo and young inflorescence were cultured on MS medium which was treated with plant growth regulators. Furthermore, the regeneration ability of callus type was investigated by cellular characteristics and enzymatic activity.
Young inflorescence had higher ability for the callus induction and plant regeneration than other somatic tissues. The combination of 2,4-D 2mg/ℓ and Kinetin 2mg/ℓ was the most effective for the callus induction and 2,4-D treated in the callus induction medium was better than NAA on the plant regeneration.
In appearance, embryogenic callus was white to pale yellow, compact, separative and knobby. Non-embryogenic callus was yellow to brownish, wet and soft. The treatment of MS medium added with 2,4-D 2mg/ℓ alone was best for the formation of embryogenic callus. In spite of increasing callus ages embryogenic callus kept its regenerative potential for a long time compared with non-embryogenic callus.
Enbryogenic callus was characterized by the dense distribution of cells, the heavy deposition of starch granules and cytoplasmic materials, and the high content of soluble protein.
