Woojeon is a green tea plucked in early to mid-April, which has a beautiful, smooth and concentrated flavor that lingers in the mouth and evaporates on the mid-tongue. Polyphenols, the main components of green tea, are antioxidant phytochemicals and are known to have protective effects against various diseases. The objective of this study is to investigate the antioxidant and anticancer activities of Woojeon on HCT116 human colon cancer cell lines.
It was found that the total polyphenol contents of Woojeon was 30.09 mg GAE/g, and antioxidant activities were 73.85%, 33.26% and 48.52 mg TEAC/g, measured by DPPH, ABTS and FRAP assay, respectively. The treatment of HCT116 human colon cancer cell lines with Woojeon at various concentrations resulted in a sequence of changed characteristic of apoptosis, including the loss of cell viability and morphological changes. Further, annexin V-fluorescein isothiocyanate and propidium iodide double staining through flow cytometry analysis demonstrated that the degree of apoptosis was increased by Woojeon. In addition, the Bax and Bcl-2 expression levels were increased in a dose-dependent manner, resulting in the activation of caspase-9, caspase-3 and cleaved poly-ADP-ribose polymerase; these observations are hallmarks of apoptotic events. Moreover, c-Jun N-terminal kinase, p38 MAPK and extracellular signal-regulated kinase was indicated Woojeon-induced apoptosis, and was accompanied by phosphorylation of the mitogen-activated protein kinase family.
