For the production of potent chitinolytic enzyme from bacteria. screening was carried out. Of 100 samples from soil of Kyung-gi and Chung-cheong area. 7 strains of chitinolytic bacteria were isolated. Among them KM-3 was identified as Pseudomonas acidovorans KM-3 which showed the higher chitinolytic activity than others. Culture conditions of Pseudomonas acidovorans KM-3 for the production of chitinolytic enzyme were inverstigated and the enzyme was partially purified by the ammonium sulfate fractionation. Maximum production of chitinolytic enzyme was obtained at pH 7.0 and 30℃ with chitin concentration between 0.2% and 1.0%. Conditions for the enzyme production were optimized using Jar-fermentor. Maximum enzyme activity was shown as 4.25 units/ml at the 24hours cultivation. Optimum temperature and pH of crude enzyme was 40℃ and pH 7.0. respectively. It could be applied to waste-treatment for the chitinous substrates.
