영문 초록

Objectives This study was conducted using the RAW 264.7 macrophage cell line to investigate how bee venom (BV) affects NALP3 inflammasomes and other inflammatory mediators in gouty inflammatory responses induced by monosodium urate (MSU) crystals. Methods To establish the model, cell viability was evaluated by WST assay, and MSU crystal was applied for each concentration to confirm the expression change of the indicators. To investigate the relationship with the MAPK pathway, 1μM and 10μM of p38, JNK, and MEK inhibitors were used as controls. MSU 1.0 mg/mL, BV 0.5, 1.0, and 5.0 μg/mL were used. RT-PCR, ELISA, and NO assay were performed to confirm the anti-inflammatory effects of BV. Results BV showed viability of more than 75% even at a concentration of 10 μM. BV suppressed NALP3, IL-1β, TNF-α, and NF-κB mRNA expression. BV inhibited the production of TNF-α, IL-6, and NO. Conclusions Our results suggest that BV could be considered as candidates for treating gouty inflammation by ameliorating inflammatory mediators.
